Growth Promotion Test

1.0 PURPOSE :
To give procedure for growth promotion test.
2.0 SCOPE :
This is applicable to check the nutritive and selective properties of media used for all microbiological analysis.
3.0 RESPONSIBILITY :
Microbiologist.
4.0 FREQUENCY :
Each lot of media prepared or must be performed atleast on each lot of dehydrated media.
5.0 PROCEDURE :
Growth promotion test is done to test each lot of the medium for its growth promoting qualities. Media plates or broth should be inoculated with relevant organisms of each strain.
The test media are satisfactory, if clear evidence of the growth appears in all inoculated media within the given incubation period.
If freshly prepared media are not used with in 2 days store them in dark at room temperature.
5.1. MEDIA PREPARATION :
Media preparation and composition refer the SOP No. QC/MB/
5.1.1 Media controls :
Each batch of medium sterilized must be checked to ensure growth of the relevant microorganism(s) and to ensure that the blank, plates are not contaminated.
5.2 TEST MICROORGANISMS


5.4 INCUBATION CONDITION & PLATE DIVISION FOR TESTING
The plates should be incubated as follows & each of the plate should be divided as follows for growth promotion testing and acts as a negative control.



5.5 BROTH PREPARATION
Dissolve in accordance with instruction on the container
the specified amount of media in required amount of distilled water and mix thoroughly. Pour the required quantity in the test tubes and sterilize by autoclaving at 15lbs (121°c) for 20 min.
5.5.1 Broth Controls
The broths should also be checked to ensure (1)sterility (2)ability to support the relevant microorganism (s).



5.6 INCUBATION CONDITIONS
Take 1 tube of each as a negative control. Incubate the other with a loop full of the test organism. Incubate each type of broth as follow.



5.7 RECORDING OF RESULTS
Result should be recorded according to the following format in a log book
